چکیده

آسیب استخوانی یکی از چالش­ های علم پزشکی محسوب می­ شود که هر ساله هزینه زیادی را در دنیا برای درمان به خود اختصاص داده است. امروزه، ساخت داربست مناسب به منظور جایگزینی برای بافت از دست رفته با هدف بازسازی، ترمیم و بازگرداندن عملکرد بافت، جهت درمان آسیب استخوانی به طورگسترده­ای مورد بررسی قرار گرفته است. ترکیب پروتئین-پلی ساکارید در داربست­ ها با قابلیت ترغیب تشکیل آپاتیت استخوانی یک روش قابل قبول برای شبیه­ سازی محیط خارج سلولی استخوانی شده است. در این مقاله، داربست­ های جدید کامپوزیتی زیست فعال، بر پایه پلی ساکارید کاپا-کاراگینان و نانوالیاف فیبرویین به منظور استفاده در کاربردهای بازسازی استخوانی، سنتز و مشخصه­ یابی شده است. نانو الیاف فیبرویین با روش الکتروریسی تهیه و به عنوان فاز تقویت کننده در مقادیر مختلف به محلول کاپا-کاراگینان 5/0 % وزنی اضافه شده و داربست­ های سه بعدی توسط فریز-درایر ساخته شده­ اند. جهت ارزیابی داربست پیشنهادی، روش­ های مشخصه یابی مختلفی به منظور آنالیز ساختاری، مورفولوژی، مکانیکی و میزان آپاتیت تشکیل شده بر روی سطح داربست­ها، به کار گرفته شده است و با نمونه کاپا-کاراگینان خالص، مورد مقایسه قرار گرفته ­اند. نتایج حاصل از میکروسکوپ الکترونی روبشی وجود ساختار متخلخل با ارتباط درونی و درصد تخلخل و میانگین اندازه حفرات مناسب با سلول­های استخوانی را نشان داده است. علاوه بر این، نتایج آنالیز طیف سنجی پراکندگی انرژی پرتو ایکس از داربست­ های قرار گرفته در محلول شبیه ­سازی شده بدن، تشکیل لایه­ ای از کلسیم فسفات بر روی سطح داربست ­ها را نشان داده است. هم چنین نتایج حاصل از آزمون اندازه­ گیری میزان سمیت سلولی، زنده ­مانی بالای 90% برای تمامی داربست­ ها، زیست­ سازگاری و عدم سمیت داربست­ ها را به اثبات رسانده است و چسبندگی و لنگراندازی مناسب سلول­ها بر روی سطح داربست­ ها که لایه ­ای سلولی ایجاد کرده اند، مشاهده گردیده است.

چکیده [English]

 Bone defect is one of the important challenges of medical science. Today, the synthesis of a suitable
scaffold has been developed. In this paper, new bioactive composite scaffolds based on kappa-carrageenan/fibrobin
nanofibers were synthesized and characterized for bone regeneration applications. The fibroin nanofibers were prepared
by electrospinning, then the chopped electrospun nanofibers were incorporated to Kappa-carrageenan solution (0.5%
w/v) in different ratios as a reinforcer. Finally, 3D scaffolds were fabricated by Freeze-drying. The results of scanning
electron microscopy showed the highly porous structure with the interconnected porous. The porosity percentage and
average pore size of the scaffolds were suitable for bone cells implantation and propagation. In addition, Energy
Dispersive X ray (EDS) technique has proved
Precipitation of Ca-P on the surfaces. The biocompatibility evaluation of
fabricated scaffolds was done using MTT indirect assay.
